These modular semi-preparative HPLC systems set standards in the purification of many samples such as peptides.
The successful semi-preparative purification is ensured by previous testing and optimization of the separation procedure if necessary. All of this with low substance and solvent consumption of an analytical HPLC. The user-configurable liquid handling system, working as autosampler and fraction collector, offers the flexibility to purify various samples within a very short time range.
Each semi-preparative HPLC is controlled by very powerful software. Besides clearly arranged method development tools and chromatogram processing functions, complete data documentation according to GLP guidelines is included.

The semi-preparative HPLC mainly fulfills two functions in the R&D laboratory. Firstly, there is a link in the upscaling from analytical chromatographic methods to preparative demands (small-scale production). It serves as a tool for determining system parameters for upscaling into the preparative scale, but secondly it is also suitable for purifying smaller amounts of substance (up to a few grams) following analytical identification. The semi-preparative HPLC is an extended version of the analytical HPLC and assures a successful transition into the preparative scale.
The semi-preparative HPLC is mainly composed of one to two HPLC pumps, one UV detector, autosampler for sample application and HPLC software for data analysis and control of the semi-preparative HPLC.
The main task of the semi-preparative HPLC is evaluating the capacity of the separation column in order to obtain the desired product in an economically sensible manner by determining parameters such as flow, pressure, load, solvent selection and separation time beforehand. These parameters add up during preparative work and will be effective production costs.
Furthermore, the semi-preparative HPLC is used to purify certain substances amounts. Therefor parameters of the analytical method are extrapolated with a so-called upscale factor, which is calculated by the column volume of analytical to semi-preparative column, followed by observation of the peak behavior (substance characteristics) on the semi-preparative column. By means of semi-preparative HPLC analytical parameters can be adapted to the preparative scale.
Established application areas for the semi-preparative HPLC are for example the production of milligram to gram quantities of metabolites in the pharmaceutical industry for further characterization by other methods, the small-scale purification of standards from natural substances or the determination of upscaling parameters for production as mentioned earlier.
Typical Modules of Semi-Preparative HPLC:
Eluent tray with tub and holder for four solvent bottles ensures a tidy system setup (bottles included).
HPLC pump with double-pistons for guaranteed accurate flows of up to 10 ml / min and system pressures of up to 400 bar. They are available as isocratic pumps or quaternary low-pressure gradient pumps (LPG) as well as binary high-pressure gradient pump (HPG) with high-pressure mixer. The high-pressure gradient pumps are equipped with two double-pistons.
Degassing: If necessary 2- and 4-channel degasser are available.
HPLC Separation Column
Porous solid particles with a large polar surface such as silica gel or alumina are suitable as solid phase. Most commonly normal and RP phases with silica particles with diameters in the micrometer range are used.
UV-Vis detector with measuring range of 190-800 nm for analytical and semi-preparative applications. Low baseline-to-noise ratio ensuring low detection thresholds and high linearity for reproducible results. The detector can be controlled either manually or via software on the control panel. The flow cell is mounted on the front and is easily accessible; the side-mounted lamps are easily replaceable.
The fraction collector provides full flexibility which is otherwise only possible with complex robotic systems. Thus, this fraction collector can be adjusted quickly and easily for the usage of any desired rack or bottle size. Minimum dead volume prevents carryover and ensures highest purity. A height-adjustable valve with practical quick lock along with specially developed software for preparative chromatography control facilitate daily work.
HPLC software for easy processing of all semi-preparative HPLC tasks considering GLP guidelines. To make all modules quickly and efficiently accessible for the operator an additional component was designed: The Navigator. It can be used for carrying out all actions by simple mouse operation via drag & drop.
