For the production of high quality HPLC columns a special packaging technique must be applied to compress the column material in the solid column tube made of stainless steel thus forming a homogeneous column bed. Only then the self-packed preparative column can be used for high pressure separation processes and provide distinct peaks after passing the HPLC detector.
For packing, the column material first is mixed in a container with a suitable solvent to an emulsion, referred to as slurry. The slurry must be stirred well, because only then a homogeneous column bed can be formed during subsequent compression in the column tube. The slurry is immediately filled into the column tube preventing it from settling down in the mixing vessel.
Now, the column piston is put into the column tube. During subsequent compression of the slurry the excess solvent escapes through the column outlet. Shortly afterwards, the specified packing pressure is reached and the pressing process is completed. The packing pressure must be significantly above the following operating pressure in the HPLC run.
The optimally designed distribution plate and the frit at the movable piston, guarantee a uniform distribution of the substance over the entire column cross-section resulting in optimal peaks.
For HPLC column usage with dynamic axial compression (DAC) the column remains in the column packing station and the column bed is automatically kept under pressure by the column piston. This prevents dead volume at the column inlet thus prolonging the life span of the preparative separation column massively whilst reducing ongoing operating costs at the same time.

A column packing station operated with static axial compression (SAC) makes manual repressing after prolonged use necessary to obtain a homogeneous column bed again. The HPLC column may be removed from both the DAC and the SAC column packing station e.g. for using the column separately. This way many columns can be prepared with a single packing station.
As previously mentioned, different column bed lengths can be packed with the column packing station which can contribute to cost savings for solvent. As an option temperature control over the full bed length is possible by a thermal jacket.
For HPLC columns with a diameter > 10 cm or in hygiene zones a slurry tank is used for the large amounts of column material and solvent. In the slurry tank, the slurry is mixed and pumped into the column tube located in the column packing station. This is, especially in the case of a large column volume, considerably easier for the user and can also be used in Ex zones.
